What’s better than a drug delivery system that truly delivers on multiple levels? Mini-tablets are a patient-centric dosage form with versatile applications. They offer a more accurate dose delivery, and their ease of ingestion makes them ideal for children, the elderly, and patients who have difficulty swallowing. This provides a degree of safety and convenience of administration. Such convenience translates into improved patient compliance, and that’s good for everybody.
For manufacturers, mini-tablets offer a highly versatile way for drug delivery wherein tablets with different release profiles can be dosed together to achieve the desired effect. This controlled drug delivery system can reduce the frequency of dosing and increase the effectiveness of the drug by localization. Mini-tablets with immediate release, delayed release and/or extended release profiles can be dosed in one capsule to achieve targeted drug delivery. They can be formulated as floating devices, for example, to enable retention in the stomach for complete drug delivery. Mini-tablets can be used to deliver two actives together, which may be especially useful if the two actives are incompatible with each other. The small size of the tablets means faster disintegration times and, thus, they can be formulated as orally disintegrating tablets.

Metrics has more than 10 years of formulation development experience making and manufacturing mini-tablets. As therapeutic benefits have made this solid dosage form design a popular trend, we have seen an increase in requests from our clients. In fact, one client requires a batch size of 20 kilos and orders four to six batches every year. Mini-tablets are easy to manufacture since they often can be formulated with a direct compression or dry granulation process and, in addition, lend themselves to a high throughput compression process using multi-tipped tooling. They also present fewer stability problems than liquid formulations which may require solvents for manufacture. Mini-tablets require less coating and offer greater flexibility during their formulation development. Overall, they are a unique dosage form which allow for a large API load to be delivered in a small tablet volume.
Mini-tablets typically have a diameter of 4mm or less and can be a tiny as 1mm. 2.1mm is the smallest size produced at Metrics today. Their size uniformity, shape and mechanical strength allow them to be filled into a capsule, in a sachet, or occasionally, further compressed into larger tablets. They also can be stick packed, and while Metrics does not offer stick pack filling and packaging, we do manufacture product for clients who favor that packaging method.
At Metrics, we compress mini-tablets using multi-tipped tooling to produce a greater number of tablets simultaneously and to save production time. A smaller tablet press is employed to afford better weight control. The mini-tablets can then be coated using a perforated coating pan or a fluid bed apparatus. We have experience with the coating of immediate release, enteric coated and modified release mini-tablets as small as 2.1mm diameter. We use mesh-lined O’Hara Technologies coating pans that allow the coating of the mini-tablets in a coating pan.
